The Fastest and Most Flexible Color Fluorescence and FISH Imaging System

Isis SystemThe Isis color fluorescence and FISH imaging system is widely recognized as the fastest and most flexible system of its kind. Isis combines ease of use and a functionality, that meets the requirements of even the most demanding research applications. With the help of automatic integration time adjustment, and with the support of many motorized microscopes, it reliably acquires fluorescence images with up to 12 color channels. For image processing Isis provides a variety of tools to enhance, edit, annotate, archive, measure, and print the images.

The optimized user interface guarantees rapid results in research and routine. Isis can be either installed as a stand-alone system, or it can be seamlessly integrated into Ikaros karyotyping stations, the Metafer slide scanning workstation, or any multi-user workplace with networked stations.

Options for color karyotyping, comparative genomic hybridization (CGH), multicolor FISH (mFISH), multicolor fluorochrome banding (mBAND), and for automatic telomere measurement are available. Like the Ikaros karyotyping software, Isis provides an integrated case database and various tools to manage and evaluate the data. Being part of the software package, the archiving software MetaArchive tracks all the work and facilitates secure and complete archiving of data and images to DVD-RAMs.

Image Acquisition and Processing

Professional imaging of FISH samples requires images of highest quality. The unique combination of state-of-the-art image acquisition hardware and software tools for automated adjustment of imaging parameters provides a stable environment for reliable microscopy imaging. Image processing with Isis is easy, yet powerful. Many image enhancement tools can also be used automatically, thus, matching the requirements of routine diagnostics and providing a reproducible workflow.

  • MetaSystems CoolCube camera
    The high resolution 1.3 megapixel USB camera is compatible with all MetaSystems imaging products. It is particularly suited to low light level situations and fluorescence imaging due to its very low power consumption and optimized housing design that keeps the imaging sensor cool, leading to a low thermal noise ratio (12-Bit, 15 fps, 1360x1024 resolution, 2/3" sensor).
  • Focusing aid
    A focusing aid with best-focus acquisition facilitates easy acquisition of images being perfectly in focus.
  • Automatic focus stack function (Z-stack images)
    The focus stack function is a simple and effective tool to analyze 3D objects. Captured images from different focus planes can be combined generating an extended focus image. All signals are clearly detectable, while unspecific background is removed. The focus stack image acquisition takes advantage of the motorized focus of many microscope models, but may also be used manually, if a non-motorized microscope is used.
  • Color transmitted light images
    The optional RGB LED illumination with adjustable light levels provides fast capture of high quality Brightfield images.
  • Automatic image processing after capture
    User-defined automatic processing functions significantly simplify the image enhancement right after capture.
  • Sophisticated tools for image enhancement
    Powerful filtering algorithms (lower and upper threshold, adjustable enhancement filter, histogram equalization...), which are applicable at any stage of image processing, generate perfectly banded chromosomes or more consistent distribution of FISH spots from even poor images.

Measurement and Analysis

Many assays require detailed analysis of image features. With its unique image display modes Isis provides full access to any detail within the image. Any time the original image can be displayed, color channels can be analyzed separately, and several adjustable false-color display modes are at hand. Various measurement tools can be used to further analyze the images. With the color karyotyping upgrade precise analysis of FISH stained chromosomes is possible.

  • Rapid chromosome classification and counting in metaphase
    Chromosome assignment in the metaphase, automated object counting, and fast classification of chromosomes is available with the color karyotyping module.
  • Various measurement possibilities
    Isis offers various measurement possibilities, including area measurement, distance measurement, and even the extraction of fluorochrome intensity profiles.
  • Multiple display modes
    Multiple customizable color display modes (RGB, individual colors, modified colors, merged colors, gray scale, inverted gray scale, combinatorial false colors) allow detailed analysis of images. All false color lookup tables can be easily modified and saved separately for further usage. 
  • View images 
    The View Images function provides a palette of tools for easy and convenient presentation of images, or for the extraction of image parts for presentation purposes. View images can be resized, rotated, freely arranged on the screen, combined, exported, copied to the clipboard, and saved as an separate Isis image. Additionally any image or karyogram can be automatically split into a set of view images representing the individual color channels.

Reporting and Data Management

Any data obtained with Isis is stored and managed in a very convenient way. Images are automatically stored together with case data, patient data, and measurement results. A powerful reporting interface allows to compile data and images to clearly arranged, customizable reports, which can be either printed or exported to PDF files. With MetaServer and MetaClient all data is indexed, and can be used to quickly generate meaningful statistical reports. MetaArchive is automatically supervising the data stock and archives all new images and cases to secure DVD-RAM data storage media.

  • Comprehensive case data management system
    Case data, images, and other files can easily be managed with the integrated Case Data Management System. Customizable data sheets with searchable fields allow fast navigation and data retrieval. Flexible automated data import and export facilitate the exchange of results with external case data management systems.
  • Powerful and flexible report generator
    Result reports based on user defined layout templates can be printed. Reports may include images, text, and the content of any case or image data field. All reports can be easily printed, exported as PDF file, or even transferred to standard word processing software. 
  • Convenient and safe archiving of images and case data
    The archiving software MetaArchive (included) offers the most convenient and safest procedures for daily incremental archiving of patient data and images, as well as third party files, to an external archiving media. All new and modified files are automatically identified and archived. Files that have been already archived are not copied again. The original directory structure is replicated on the archiving media, making it easy to locate any archived case.
  • Multi Data Retrieval and Search Possibilities
    The convenient MetaServer / MetaClient software bundle is an integral part of every Ikaros and Isis installation. MetaServer and MetaClient use background processes to automatically collect, sort, and display data generated by all Ikaros or Isis workstations within a network or on a stand alone system. Multiple data retrieval and search possibilities allow for easiest handling of even complex patient databases.

Configuration and Updates

    Individual user accounts allow multiple user accessing the same set of data at the same time. The built-in networking capability allows flexible multi-user environments combining complete workstations with capture, analysis or review stations.

    Multiple images from the same case can be cooperatively processed at the same time from different locations, enhancing significantly the efficiency.  All processing steps are updated immediately on the central server and are available on all clients without the least delay.
