Isis - mFISH and mBAND Software
XCyting Colors - Complex Aberrations
The software upgrade for multicolor FISH (mFISH) and multicolor chromosome banding (mBAND), available as an extension for the fluorescence imaging system Isis, provides the tools to analyze the chromosomal origin of each individual pixel within an mFISH or mBAND image. Thus, marker chromosomes, complex chromosomal rearrangements, and all numerical aberrations can be visualized simultaneously in a single hybridization experiment. By analyzing the color ratio of of all pixels, the software unambiguously assigns any region of the image to the respective chomosome class. The filter based mFISH approach facilitates a direct inspection of the original hybridization signals and, thus, overcomes the need for additional control experiments with conventional chromosomal paints.
MetaSystems' proprietary high resolution multicolor banding technique mBAND provides precise information on intrachromosomal rearrangements and exact break-point localization. mBAND probe kits comprise overlapping partial chromosome paints, which resemble a reproducible false-color banding pattern, when visualized in the mFISH/mBAND software. mBAND patterns are independent of chromosome condensation, and are easy to analyze.
The Isis color karyotyping software upgrade is required for mFISH and mBAND with Isis.
Multicolor FISH (mFISH)
MetaSystems provides an intergrated system-plus-reagents solution for multicolor chromosome analysis. The 24XCyte mFISH probe kit consists of a set of chromosome painting probes with specific fluorochrome combinations covering the human genome. mFISH kits are also available for mouse, rat, and Chinese hamster ovary (CHO) chromosomes (21XMouse, 22XRat, 12XCHamster).

The Classification Tolerance and Minimal Classification Intensity tools optimize the color classification in mFISH and mBAND by excluding pixels with ambiguous color signatures from the classification. This way, regions with bad hybridization or artifacts are easily identified and suppressed. - Single color gallery
The Single Color Gallery is a separate window in which all five color channels are displayed for a selected chromosome. Additionally the false color image, an inverted DAPI image, and the fluorochrome profiles are shown. With the help of the Single Color Gallery the identification of rearrangements is extremely easy and straightforward.
Multicolor Chromosome Banding (mBAND)
For mBAND MetaSystems provides the XCyte mBAND probe kit, which is available for all human chromosome classes.
- Slicing and color ratio analysis
Each XCyte mBand probe kit consists of overlapping partial chromosome paints. The Isis mBAND software virtually cuts the chromosome into artificial bands and assigns pseudo colors to them, based on the unique color combination of each slice. - Adaptable mBAND resolution
The resolution of the mBAND pattern can be modulated in order to get the most suitable image for analysis. A resolution equivalent to 550-band level for G-bands can be easily archieved. mBAND patterns are independent of chromatin condensation. - Direct correlation to ISCN bands
Though mBAND bands resemble an artificial banding pattern, they can be directly correlated to ISCN bands with the help of the single color gallery. This way precise breakpoint localization is possible.
