The telomere module is available as an upgrade for Isis. Telomeres of chromosomes labelled with a common telomere FISH probe can be automatically analyzed. Quantifying the amount of telomeric DNA in cancer specimens is an area of growing interest in cytogenetics, since telomeres are essential for the stability and the replication of chromosomes. Telomeres are also thought to play an important role in senescence and apoptosis.
The Isis color karyotyping software upgrade is required for telomere analysis with Isis.
- Automatic detection of telomere signals
Telomere signals are automatically identified and analyzed for their intensity.
- Measurement regions
The measurement regions can be dynamically set with the mouse.
- Reference signals
Reference signals such as centromere probes can be measured in the same color channel like the telomere signals, or using a different fluorochrome. The telomere data can be normalized, dividing it by the reference signal intensity. With this strategy variations in hybridization efficiency between individual metaphases can be compensated.
- Background correction
The intensity value for each pixel is corrected for the background intensity, and during the measurement the intensity is summarized for the telomere area.
- Displaying the results
Results from the telomere measurement are automatically saved into text files. They can be displayed on screen, and are summarized for all karyotypes that have been analyzed. Data are displayed either as tables or graphically, and are shown as raw data or normalized. Statistics summarize the results of different karyotypes. Images and figures can be exported to standard file formats.
