Automated Imaging

Each microscopic sample is different; automation of imaging therefore demands flexibility, but is exempt from subjectivity. Metafer's unique concept provides a subtle solution: a user-trainable classifier precisely defines the scanning standards; and with the selection of another classifier the system is quickly switched to the next task. This is the reason for MetaCyte's unrivalled flexibility.

Interphase FISH

The analysis of spot signal patterns is the basis for many assays in hematology and cancer genetics. With MetaCyte, based on the Metafer platform, these patterns are analyzed automatically, precisely, and reproducibly. Results are stored together with nuclei images and can be displayed in the image gallery or in convenient histograms and scatter plots. Data can be summarized in customizable reports for printing or exporting. With a few mouse clicks, a selective display of cell sub-populations based on their spot patterns (gating) is possible. In combination with the high-quality XL DNA probe kits for locus specific hybridizations, MetaCyte becomes an all-encompassing unit for professional routine diagnostics.

  • Fully automated detection of cells and / or nuclei
    Morphology and labeling parameter can be set in the classifier to precisely define the target cells.
  • Image enhancement
    Images can be automatically enhanced based on a variety of algorithms. Color channels, analysis images, and gallery images are treated individually.
  • Analysis of cell features
    Spots are analyzed individually for each color channel. Signal and / or fusion counts are displayed in the gallery of nuclei. Signal patterns can be summarized, and classes with subpopulations based on spot and fusion counts are automatically generated and displayed.
  • Gating
    Cells can be selected based on any combination of obtained features. Convenient histograms and / or scatter plots provide a quick overview over the cell and feature distribution, and populations are selected with a few clicks of the mouse. Target cells can be selectively displayed; however, the total amount of data remains untouched, and even hidden data can always be retrieved.
  • Reporting
    All results can be easily compiled into customizable reports. Raw data are always available for export.

Tissue Imaging

The analysis of tissue sections requires clearly defined strategies to identify regions of interest. The revolutionary Tile Sampling algorithm automatically detects regions with defined staining patterns in situations where segmentation of individual cells is impossible. Pre-scan options provide a sample overview. And a convenient tool to edit tissue micro array (TMA) patterns facilitates high-throughput analyses. Due to this functionality, Metafer has been successfully applied to diagnosis of solid tumors, no matter whether based on conventional tissue sections or on TMA preparation.

  • Tile Sampling
    Automated detection of regions for analysis in tissue sections, based on the revolutionary Tile Sampling algorithm.
  • Individual focussing
    Extended focus images are acquired automatically by MetaCyte. Classifier settings define individually for each color channel how many focus planes are captured.
  • Tissue microarray (TMA) functionality
    The tissue microarray tool, which is part of all MetaCyte systems, facilitates the definition and read-out of TMA samples. Pre-scans at low power magnification quickly provide a preview of the sample, and core centers are automatically detected. The position list generated from the pre-scan can directly be used for the analysis at high magnification.
  • Amplification analysis
    Based on MetaCyte's range of image analysis parameters, amplification states of cells in a tissue can be analyzed automatically. MetaSystems offers a unique system for automated Her2/neu breast cancer analysis.

MetaCyte, upgraded with VSlide, creates accurately stitched brightfield or fluorescence overview images while scanning the slide. True color images from brightfield illuminated specimen are generated either with the CoolCube 1c color camera, or with the CoolCube 1m monochrome camera, using an integrated, fast and accurate color LED illumination.
